**Disclaimer:** Constant-temperature exposure only. Real cycles integrate lethality over the heat-up and cool-down profile from calibrated probes. Cycle acceptance follows your validated procedure.

## Content

**Intro:** F0 expresses the lethality of a moist heat cycle as equivalent minutes at 121.1 C. It is used to compare cycles, set minimum exposure for loads and prove that a biological indicator was inactivated by enough log cycles.

**Sections:**

#### How to use it

**Body:** Enter the temperature and the time held at it, then keep z at 10 unless your validation uses another value. Add the D value from the biological indicator certificate to see the log reduction. Set a target F0 to see the shortfall and the time that would be needed at this temperature. For a full cycle, break the profile into short intervals and add up the F0 of each.

#### Formula and worked example

**Body:** Lethality rate equals 10 raised to (T minus 121.1) divided by z. F0 equals time multiplied by that rate. At 121 C for 15 minutes the rate is 0.977 and F0 is about 14.7 minutes. At 118 C the rate drops to roughly 0.49, so the same 15 minutes only gives about 7.3. A temperature drop of 10 C cuts lethality by a factor of ten.

#### Common mistakes

**Body:** Averaging temperatures before applying the formula understates or overstates lethality, because the relationship is exponential. Using the chamber drain probe instead of a coldest-spot load probe is another classic error. Also check that F0 is not used as the only criterion for loads where bioburden resistance is unknown.

**Cta:** Netray can bring cycle records, equipment IDs and load release data from your ERP and MES together on-prem. Ask for an expert review.

**Faq:**

#### Why is the reference temperature 121.1 C?

**Answer:** It converts the historical 250 F reference into Celsius. The convention keeps F0 values comparable between sites and the literature.

#### What F0 is enough?

**Answer:** It depends on bioburden, indicator resistance and the target sterility assurance level. Many porous and liquid cycles target F0 of at least 15 minutes, but your validation decides.

#### What does the D value mean?

**Answer:** It is the time at a given temperature to reduce the population of a microorganism by 90 percent, or one log. More log reduction needs more time.

#### Can I use this for dry heat?

**Answer:** Dry heat uses a different reference temperature and z value, usually described as FH. Change the inputs only if your validated method specifies it.
