Endotoxin Limit and Maximum Valid Dilution (MVD) Calculator

Every sterile product needs a justified endotoxin limit and an MVD that lets the LAL test see it. This calculator applies the K over M method and gives MVD so you can plan product dilution and inhibition and enhancement testing.

Your numbers

kg

70 kg is the usual adult basis; use a lower value for pediatric products if required.

mL/h

Largest volume given per hour (or per dose for single-dose limits).

EU/mL

1 for liquid products tested as is. For powders use the reconstituted concentration ratio.

How to use it

Select the threshold pyrogenic dose that applies to the route, enter the body weight basis and the largest dose volume per hour, then add the assay sensitivity from your reagent certificate. The result is the endotoxin limit in EU/mL and the maximum dilution you may use. A product that needs a bigger dilution than the MVD to remove interference is a method development issue.

Formulas

The limit equals K divided by M, where M is the maximum dose per kg per hour. With K = 5 EU/kg, a 70 kg person and 10 mL per hour the limit is 35 EU/mL. MVD equals the endotoxin limit times the concentration factor divided by lambda. With lambda 0.03 EU/mL the MVD is about 1167, enough for roughly ten two-fold dilutions.

  • EL = K x W / dose volume
  • MVD = EL x c / lambda
  • Dilutions are limited by MVD

Common mistakes

Forgetting that the intrathecal threshold is per dose and not per hour, mixing units between mL per hour and mL per kg, and using a lambda that is not the labeled sensitivity of the lot of reagent in use are common. Also remember that a high MVD does not guarantee absence of inhibition, so product-specific validation is still needed.

Frequently Asked Questions

What does K mean?

K is the threshold pyrogenic dose, the amount of endotoxin per kg that can cause a fever response in people. It is 5 EU/kg/h for most parenteral products and 0.2 EU/kg for intrathecal use.

What is MVD used for?

It sets the highest dilution at which a sample can be tested while still detecting endotoxin at the limit. It guides the dilution used to overcome product interference.

Which volume should I enter?

Use the maximum dose volume in the labeled use, expressed per hour for most products. For single-dose products with a defined duration, follow your approved calculation.

Is this valid for devices?

Device limits are normally expressed per device or per extraction volume and use different rules, so do not apply this form directly.
